    About The Presenter

    J. Harry Sweigard, PhD
    Principal Scientist , Absorption

    Dr. Harry Sweigard is a Principal Scientist in the Ophthalmology unit at Absorption Systems in San Diego California.  Prior to joining Absorption Systems he was a lab head at Novartis Institutes for Biomedical Research where he studied the underlying mechanisms of ocular inflammation.  During his postdoctoral training and as an instructor at Massachusetts Eye and Ear Infirmary/Harvard Medical School he used a diverse set of rodent models of vascular retinopathy to better understand innate immune system involvement in retinal vascular homeostasis.  This work led to several peer-reviewed articles including a recent publication in Science Translational Medicine. This was a natural segue from his doctoral work at Tufts Sackler School where he designed novel viral vectors aimed at dampening innate immune system activation towards therapies for age-related macular degeneration.

    Dr Sweigard applies his extensive experience and expertise in ocular and neurodegenerative disease modeling to develop and validate in vivo models at Absorption Systems that can be used to determine efficacy, distribution and safety of novel ocular therapies and delivery systems.

    A workshop that examines the challenges with establishing bioequivalence of locally acting topical, ocular, and inhalation pharmaceutical products and which presents various relevant solutions that could form the basis of best practice approaches for evaluating the bioequivalence of such products.

    Held in conjunction with the 2016 AAPS Annual Meeting and Exposition.

    Background

    The demonstration of bioequivalence for locally acting drugs, for which the site of action is at or near the site of application and upstream from the systemic circulation, has frequently posed some of the greatest challenges in bioequivalence. Historically, extremely expensive clinical endpoint bioequivalence studies have generally been the centerpiece of bioequivalence assessments for such products. In addition to the high cost and difficulty associated with such clinical endpoint studies, they are notoriously insensitive toward formulation differences. Although these problems have plagued those firms seeking to develop a variety of different dosage forms, three of the most problematic routes of administration have been topical (dermatological), ocular, and inhalation. The vast majority of publications and public meetings addressing bioequivalence for products administered via these three routes have focused narrowly on a single route of administration. However, there are many challenges that are common to the development of alternative bioequivalence strategies of products administered via these three non-oral routes.

    Goals

    This workshop seeks to bring together the latest research and thinking from these three fields to explore the possibilities of developing more unified, totality of evidence-based approaches toward the demonstration of bioequivalence for these products in ways that significantly reduce the clinical burden.

    Who Should Attend

    This workshop will be of particular interest to scientists in preclinical and clinical development, interested in branded and generic small molecule therapeutic drug products. Particularly relevant for small to medium size companies who are trying to enter the space of locally acting drug products.

    Biotherapeutic products are the fastest growing medicines in the pharmaceutical market. Despite their clinical success, the immunogenicity of biotherapeutics continues to be a major concern. Assessment of immunogenicity as well as appropriate interpretation of immunogenicity data is therefore, of critical importance for defining safety profile and it is one of the regulatory requirements for the approval of these products. In the past decade, much progress has been made towards how immunogenicity should be studied.     The purpose of this lunch and learn workshop is to review the current landscape of ocular drugs, address critical delivery challenges, and highlight the utility of preclinical models and tools to promote scientific understanding and support the development of novel therapies and delivery systems for anterior and posterior ocular indications.

    Research and development in this  area has witnessed significant development in recent years, which is highlighted by the approvals of drugs for the treatment of front and back of the eye diseases. An increasing number of therapeutics including biologics, stem cells, gene therapy along with modified reformulated small molecules are entering the development pipelines. However the clinical advancement of these therapeutics, is hampered by obstacles encountered in delivery due to the special anatomy and physiology of the eye.   It is therefore critical to develop strategies to deliver these agents to ensure long–lasting ocular therapeutic effects with minimal safety concerns and ultimately ease of patient use.

    This targeted and comprehensive event will provide a good learning opportunity for all scientists in the field of ophthalmology. The attendees will bring home not only a good understanding of current state‐of‐the‐art developments in ocular drug delivery and disposition but have the additional exciting experience of learning directly from the experts.
