The human intestinal Caco-2 cell line has been used extensively as a model of the intestinal barrier. Originally obtained from a human colon adenocarcinoma, the cells undergo a process of spontaneous differentiation in culture, leading to the formation of a monolayer expressing a phenotype characteristic of mature enterocytes. Our co-founder and Chief Scientific Officer, Ismael Hidalgo, pioneered the use of Caco-2 cell monolayers to predict intestinal drug absorption twenty years ago. His seminal paper on the topic (Hidalgo et al. Gastroenterology 1989; 96:736-749) has been cited more than 1000 times.
